Induction of triploidy in Rhamdia quelen (Pisces, Pimelodidae)

Abstract

 

The triploidy induction technique in Rhamdia quelen was standardized by the application of heat shocks with different durations and in different moments post-fecundation. The survival to the heat shock was measured in the moment of larvae eclosion. In order to identify the triploid fishes, the rate of ploidy was calculated by recounting the maximum number of nucleolus in interphase nuclei. The best method to induce triploidy in Rhamdia quelen was determined based on the values of the variables analyzed in this paper. The variable that produced the greatest rate of survival to the heat shock was also indicated - temperature of the shock: 36 ± 1°C; duration of the temperature shock: 5 minutes; moment of the shock temperature: 5 minutes post-fecundation, with triploidy and survival rates of 74.8% and 33.4%, respectively.
